MacroGenics is privately-held pharmaceutical and biotechnology company that is engaged in the development of immunotherapeutic treatments for cancer, autoimmune disorders, and infectious diseases. The company is helping researchers from Washington University and the National Institutes of Health to develop a fully human monoclonal antibody treatment for the deadly West Nile virus. Through an agreement with Tolerance Therapeutics, MacroGenics has expanded its efforts to include the development of another anti-CD3 monoclonal antibody used to treat Type 1 diabetes (also known as juvenile diabetes). The company has been conducting clinical trials on its cancer treatments.
Genzyme Corporation and MacroGenics have engaged in a joint development program to develop CD16 Mab and Fc soluble receptors that may offer potential treatments for immune-mediated diseases such as rheumatoid arthritis, idiopathic thrombocytopenia purpura, lupus; as well as others.

MacroGenics has taken a lot of time to develop its product pipeline with therapeutic drug candidates, both clinical and early-stage, that present a number of possible indication in diseases like diabetes, leukemia, solid tumors, West Nile virus, Multiple Sclerosis, etc.
Autoimmunity: Anti-CD3 - The development of type 1 diabetes is believed to proceed by a two-step process. In the first step, an environmental agent, possibly a virus, injures the insulin-producing beta cells of the pancreas. It is presumed that this initial injury to the beta cells precipitates a T-lymphocyte-dependent autoimmune attack on the beta cells in the pancreas, resulting in progressive destruction of the capacity to produce insulin and the signs and symptoms of diabetes. A substantial body of data in experimental animals with diabetes and preliminary data in humans with type 1 diabetes indicate that the autoimmune attack on the pancreatic beta cells can be aborted or significantly slowed with short-term intravenous treatment with monoclonal antibodies that target the CD3 antigen expressed by T lymphocytes and genetically engineered with changes in the Fc component of antibody.
hOKT3 (ala-ala) - Macrogenics, partnering with NIH-funded Immune Tolerance Network, is developing a humanized anti-CD3 monoclonal antibody (with a modified Fc component) that possesses the capability to interfere with the autoimmune mechanism which may destroy the islet and retard the progression of Type 1 diabetes. hOKT3 may also have potential therapeutic benefits in a wide range of immunological diseases including systemic lupus, ulcerative colitis and psoriasis and is being considered for studies in these and other indications.
GMA161 Anti-CD16 (FcyRIII) - An antibiotic drug candidate and a CD16 receptor inhibitor that is based on MacroGenics understanding that the prevention of CD16/Fc’s proper function can produce an effective treatment for antibody mediated autoimmune diseases such as rheumatoid arthritis, lupus, and idiopathic thrombocytopenic purpura (ITP)
Cancer: MGA321 Anti-CD32B - An antibiotic drug candidate that targets CD32B a factor related to the activity of certain cancers, and which provides an area to concentrate cancer therapy efforts. MacroGenics is developing therapeutic antibodies that block Fc engagement of CD32B may improve the performance of other cancer Mabs when combined with them during administration. MacroGenics also believes that autoimmune and inflammatory diseases may be treated using an effective CD32B inhibitor therapy.
Infectious Diseases: West Nile Virus MGAWN1 (Anti-WNV) and MGVWN1 (WNV vaccine) - both of these vaccines are in various stages of clinical development create drug products that will be highly effective treatment agents for West Nile Virus.
